Medical Uses
Portrazza is an EGFR antagonist approved to be used in conjunction with cisplatin and gemcitabine, for first-line treatment of metastatic squamous non-small cell lung cancer (NSCLC). This medicine is not for the treatment of non-squamous non-small cell lung cancer.
Recommended Dosage: The recommended dosage is 800 mg given as an intravenous (IV) infusion over an hour on Days 1 & 8 of each 3-week cycle before the infusion of gemcitabine and cisplatin. Continue treatment with Portrazza injection until the disease is progressive or unacceptable toxicity occurs.
For those who have previously experienced Grade 1/2 infusion-related reaction (IRR), pre-medicate with diphenhydramine hydrochloride (or equivalent) before all subsequent infusions of Portrazza.
For those who have experienced a second Grade 1/2 occurrence of IRR, pre-medicate for all subsequent infusions, with acetaminophen (or equivalent), diphenhydramine hydrochloride (or equivalent), and dexamethasone (or equivalent) before each infusion of Portrazza
Warning & Precautions
- Closely monitor patients taking treatment with Portrazza for serum electrolytes, including serum magnesium, calcium, and potassium before each Portrazza infusion during treatment and after the administration of Portrazza for at least 8 weeks after the final dose.
- Hypomagnesemia may occur in those treated with Necitumumab injection. Monitor patients for hypocalcemia, hypomagnesemia, and hypokalemia before each Necitumumab infusion, during treatment, and for at least 8 weeks after the completion of Necitumumab.
- Venous and arterial thromboembolic events (VTE and ATE), some fatal, may occur with Portrazza in conjunction with cisplatin and gemcitabine. Discontinue treatment for patients with severe or life-threatening venous and arterial thromboembolic events (VTE or ATE).
- Dermatologic toxicities, including acne, rash, dry skin, dermatitis acneiform, pruritus, generalized rash, maculopapular rash, and skin fissures may occur in patients receiving Portrazza in Study 1. Discontinue treatment for Grade 4 skin reactions, or for Grade 3 skin induration/fibrosis.
- Portrazza 800 mg/50 mL (16 mg/mL) treated patients may experience infusion-related reactions (IRRs) of any severity. Monitor patients during and following the infusion of Portrazza for signs and symptoms of infusion-related reactions. Discontinue treatment for serious/life-threatening infusion-related reactions.
- Based on animal findings and its mechanism of action, Necitumumab 800 mg/50 mL (16 mg/mL) can be responsible for causing fetal harm when administered to pregnant females. Women of reproductive age should use suitable contraception during treatment and for 3 months after the final dose.
