Medical Uses
Saphnelo (anifrolumab-fnia) is a type I interferon (IFN) receptor antagonist used to treat adults with moderate to severe systemic lupus erythematosus (SLE), who are taking other lupus drugs, regardless of race or ethnicity standard therapy.
Recommended Dosage: Saphnelo is supplied as a single-dose vial. It must be diluted before intravenous administration. The recommended dosage regimen of Saphnelo injection is 300 mg, administered as an intravenous infusion over a 30-minute time span, every 4 weeks.
If a planned/scheduled infusion of Saphnelo is missed, administer it as promptly as possible. A minimum interval of 14 days between infusions should be maintained.
Warning & Precautions
- Severe and sometimes fatal infections have occurred in those with immunosuppressive agents, including Saphnelo injection. If a patient develops an infection or is not responding to standard anti-infective therapy, assess the patient precisely and interrupt treatment until the infection resolves.
- Events of angioedema, infusion-related reactions, and serious hypersensitivity reactions have been noticed following Saphnelo administration. Consider pre-medication prior to infusion for those with a known history of these reactions. If infusion-related reactions, or hypersensitivity reactions, promptly interrupt the infusion.
- The use of immunosuppressants may increase the risk of malignancies. Consider the benefit-risk in those with known risk factors for the development of reoccurrence of malignancy before using Saphnelo. In those who develop malignancies, consider the risk-benefit of continued therapy with Saphnelo.
- Saphnelo 300 mg/2 mL medication has not been evaluated in combination with other biologic therapies, including B-cell-targeted therapies. Therefore, the administration of this injection is not recommended for use along with biological therapies.
- Update immunizations, as per the current immunization guidelines, before initiating treatment with Saphnelo. Concurrent use of live or live-attenuated vaccines should be avoided in patients treated with this medicinal product.
