Medical Uses
Reblozyl is an erythroid maturation agent used for the treatment of:
- Anemia in adults with beta (β) thalassemia who require regular red blood cell (RBC) transfusions.
- Anemia failing an erythropoiesis stimulating agent and requiring two or more RBC units over 8 weeks in adults with very low- to intermediate-risk myelodysplastic syndromes with ring sideroblasts (MDS-RS) or with myelodysplastic/myeloproliferative neoplasm with ring sideroblasts and thrombocytosis (MDS/MPN-RS-T).
Limitations Of Use: This medicinal product is not for use as a substitute for RBC transfusions in those who require immediate correction of anemia.
Recommended Dosage: The recommended starting dose is 1 mg/kg once every 3 weeks by subcutaneous injection for patients with beta thalassemia. Prior to each dose of Reblozyl injection, review the patient’s hemoglobin and transfusion record. Titrate the dose based on responses.
Discontinue treatment if a patient does not experience a decrease in transfusion burden after 9 weeks of therapy (administration of 3 doses) at the maximum dose level or in case any unacceptable toxicity occurs at any time. If a scheduled or planned administration of Reblozyl is missed or delayed, administer it as soon as possible and continue dosing as prescribed, with at least three weeks between doses.
Warning & Precautions
- Assess and review hemoglobin results before each administration of luspatercept-aamt injection. In case an Red Blood Cell transfusion occurred before dosing, consider the pretransfusion hemoglobin for dose evaluation.
- In adult patients with beta thalassemia, TEE or thromboembolic events were reported in Reblozyl-treated patients. Monitor patients with Reblozyl for signs/symptoms of thromboembolic events and institute treatment promptly.
- Hypertension was noted in Reblozyl-treated patients. Blood pressure should be monitored before each administration. Manage new-onset hypertension or exacerbations of preexisting hypertension with the help of anti-hypertensive agents.
- Findings from the animal reproductive studies suggest that Reblozyl 75 mg injection may cause fetal harm if administered to a pregnant woman. Females of reproductive potential should use an effective method of contraception during therapy and for at least three months after the final dose.
