Medical Uses
Lorbrena is a kinase inhibitor used for the treatment of adult patients with metastatic non-small cell lung cancer (NSCLC) whose tumors are anaplastic lymphoma kinase (ALK)-positive.
Recommended Dosage: The recommended dosage of Lorbrena Tablets is 100 mg taken orally once daily, with or without food, until the disease is progressive or unacceptable toxicity occurs. Swallow tablets completely. It is not advisable to chew, crush or split them. Do not ingest in case the tablets are broken, cracked, or otherwise not intact.
Take this medicine persistently each day. In case a dose is missed, then take the missed dose unless the next dose is due within four hours. Do not take a couple of doses at the same time in order to make up for a missed dose. Do not take any additional dose in case vomiting occurs after dosage but continue with the very next scheduled dose.
Warning & Precautions
- A broad spectrum of CNS or central nervous system effects can emerge in patients receiving Lorbrena 25 mg. These include psychotic effects, seizures and changes in cognitive function, mood (including suicidal ideation), mental status, speech, and sleep.
- Boost in serum cholesterol and triglycerides can emerge in patients taking Lorlatinib. Initiate or boost the dose of lipid-reducing agents in those with hyperlipidemia. Assess serum cholesterol and triglycerides prior to initiating Lorlatinib, 1 and 2 months after initiating Lorlatinib, and periodically thereafter.
- PR interval prolongation and atrioventricular (AV) block can appear in patients receiving Lorlatinib. Monitor ECG before initiating Lorlatinib and periodically thereafter. Withhold and resume at a lowered dose or at the same dose in those who undergo pacemaker placement. Permanently interrupt for recurrence in those without a pacemaker.
- Severe or hazardous pulmonary adverse reactions consistent with ILD/pneumonitis can emerge with Lorlatinib. Promptly assess for ILD/pneumonitis in those who present with worsening of respiratory signs/symptoms indicative of ILD/pneumonitis (e.g., cough, dyspnea, and fever).
- Hypertension can appear in those receiving Lorlatinib 25 mg. Assess fasting serum glucose prior to starting Lorlatinib and evaluate periodically thereafter. Withhold and resume at a lowered dose or permanently interrupt treatment based on severity.
- Based on data/findings and its mechanism of action, Lorlatinib can cause fetal harm if used by a pregnant woman. Females of reproductive age or potential should use an appropriate non-hormonal way of contraception.
